Mammalia : typical blood smear
Findings
Normal neutrophil @1.1 :
Size: about 13-15 μm in diameter.
Regular and rounded shape.
They have a lobed nucleus with usually more than 4 lobes. It is not uncommon for the nucleus to be hyper segmented. The chromatin is dark violet and mottled.
Their cytoplasm is abundant, light gray, and contains fine violet granulations more or less visible.
Normal Eosinophil @2.1 :
Size: close to the size of neutrophils.
Regular and rounded shape.
They have a multi-lobed nucleus with usually 2 or 3 clearly visible lobes.
The cytoplasm is gray more or less visible and filled with pink granulations often voluminous.
Normal basophil @3.1 :
Size: close to the size of neutrophils.
Regular and rounded shape.
They have a more or less visible multi-lobed nucleus because it is covered with dark purple granules more or less bulky. These granulations are usually visible.
Light cytoplasm.
Small lymphocytes @4.1 :
they are slightly larger than red blood cells (8-10 μm in diameter).
Rounded cells with a high nucleocytoplasmic ratio close to 1.
Their nucleus is round or oval, with a dark and dense chromatin.
Their cytoplasm is scarce, dark gray-blue and without granulation.
Large lymphocytes @5.1 :
Cell with a larger size (12-14μm in diameter).
Their nucleocytoplasmic ratio is lower, their nucleus oval or elongated with less condensed chromatin than the small lymphocytes.
Often the nucleus adheres to one side of the cytoplasmic membrane.
Their cytoplasm is bluish, light and smooth, their contours are often shaded.
Lymphocytes with granules @6.1 :
Cell with the same size as large lymphocytes.
These are large lymphocytes that contain small purple cytoplasmic granules that are clearly visible and individualized.
Normal monocyte @7.1 :
Size: it is the largest leucocyte (about 18 μm in diameter).
Cell with various forms.
Nucleus irregularly shaped with folds or notches but no lobe, can be serpentiform or with a clover shape. Irregular chromatin, usually mottled.
The cytoplasm is bluish "in stormy sky" with a granular appearance, and shaded. It most often contains vacuoles.
