The pretectal nuclei are groups of neuronal cell bodies within the pretectum that receive visual and other sensory inputs and participate particularly in pupillary and visuomotor reflexes.

Olivary pretectal nucleus

The olivary pretectal nucleus is especially important in the pupillary light reflex. It receives direct retinal input through fibres associated with the optic tract. Neurons from the olivary pretectal region project bilaterally to the Edinger–Westphal nuclei, allowing illumination of one retina to produce constriction of both pupils. The pathway can be summarised as:

Retina → optic nerve → optic chiasm → optic tract → olivary pretectal nucleus → bilateral Edinger–Westphal nuclei → oculomotor nerve → ciliary ganglion → sphincter pupillae → pupillary constriction.

The bilateral pretectal projection accounts for the direct light reflex in the illuminated eye and the consensual light reflex in the opposite eye.

Nucleus of optic tract

The nucleus of the optic tract and other smaller pretectal nuclei also receive retinal input and communicate with other visual centres. They contribute particularly to optokinetic and visuomotor responses, helping coordinate reflex eye movements in response to movement of the visual field.

Anterior pretectal nucleus

The anterior pretectal nucleus is a comparatively large pretectal cell mass and differs anatomically and functionally from many of the smaller pretectal nuclei. Its superficial portion receives retinal input, whereas its deeper components have broader sensory and ascending and descending connections. Experimental evidence also suggests involvement of its deeper regions in the processing and modulation of noxious somatosensory information.

Clinical correlate

Lesions affecting the pretectal region, particularly around the posterior commissure, can disturb the pupillary light reflex and may occur as part of dorsal midbrain (Parinaud) syndrome.
